What is qPCR? 

qPCR is a detection method for infectious diseases caused by viruses, bacteria, protozoa and other pathogens, and is the gold standard for viral nucleic acid detection. qPCR testing is usually required to be done in a dedicated PCR laboratory. The test has higher accuracy, higher specificity and higher sensitivity than antigen, antibody and isothermal PCR, and is the best method for disease prevention, screening and diagnosis.

Routine procedures for nucleic acid testing:

(1) Sample collection and pre-treatment

This process is mainly used to collect blood, swab, stool, saliva, and other samples from patients and usually requires different sets for sample collection. Plasmodium, Ebola, and yellow fever virus can all be tested for downstream nucleic acid detection by extracting their nucleic acids from whole blood and serum samples.

(2) Pathogen nucleic acid extraction

This process is mainly used for the extraction of nucleic acids from the samples to be examined, and the addition of the extracted template to the PCR reaction system. Nucleic acid extraction is divided into manual extraction and instrumented automated extraction. Among them, the automatic nucleic acid purification extraction has the advantages of fast extraction speed, stability and reliability, high extraction sensitivity, and good homogeneity. (3) qPCR on-machine amplification

This step is mainly to place the prepared PCR detection system reagent into the qPCR instrument for PCR amplification reaction, and conduct qualitative detection of pathogens through the principle of PCR continuous amplification method. 

(4) Report

This step is to output detection results through fluorescence quantitative detection results data, which may involve printers and other equipment.

(5) Attention

The qPCR experimental operation should be performed strictly in accordance with the PCR partition zone required by the Ministry of Health, which is generally divided into reagent preparation zone, sample processing zone, and PCR detection zone. Items in each zone are dedicated and shall not be cross-used to avoid contamination, and the bench shall be cleaned immediately after the experiment.
